Direct after installing Powertoys it also didn't work for me. But after installing the
.net core desktop runtime 3.1.3 and rebooting it was fixed for me.
I toggled the color picker on and off once or twice, and that got it working for me.

Toggling the picker on and off worked for me as well. Weird bug

For me, the color picker stops working after locking and unlocking windows. Whenever I lock and unlock windows, I need to disable and activate the color picker using Power Tools.

Faced the same issue.
In a nutshell, these are the problems that I see now (version: v0.20.1):
The Win + Shift + C wasn't working for me, but I was getting it randomly pop up when typing and I couldn't work out what key combination was causing this.
Toggling the picker on and off a couple of times has also got the Win + Shift + C shortcut working.
I'll keep an eye on whether it continues to pop up during general typing.
It has been randomly popping out for me when typing, and what I figured out is it randomly pops up when I type C
